The answer is it depends..... on where the ship was coming from, what particular port it was headed to, whether the lists have been indexed and put on-line, and so on.

Do you have any more details? If so, I or someone else may be able to provide a better answer.

Oh, and in addition to regular passenger lists, I've found some people in that era (or just a little earlier) listed in newspaper summaries of passengers arriving. Just in name in those cases, but it told me where to search further.
McGee (Donegal to Edinburgh), Jamieson/Guthrie (Leith), Keddie (Peebles, Galashiels), Little (Cavers, Traquair), Arthur (Galashiels) , Paterson (Edinburgh, with occ. spells in Stirling, Greenock, Leith), Ralston (Glasgow to Stirling), Greig (Elgin)

Be thankful that your people were not on the Annie Jane which left Liverpool in 1853, and sunk in a storm amongst the Scottish islands losing around three hundred passengers and crew.

Hi Jan:

Castle Garden was the immigration centre for the United States before Ellis Island. http://www.castlegarden.org/

You can also check passenger lists at: http://www.theshipslist.com/

Castle Garden was the immigration center for only New York City.

Records for arrivals at other ports (Boston, Philadelphia, and Savannah were three other big ones in the mid 1800s) are indexed separately, so you'll need to look at those if the people you are looking for didn't come through NYC.
